The Department of Pediatrics, Cellular and Molecular Therapy is seeking intermediate level scientists with experience in gene therapy research, specifically adeno-associated viruses (AAV), for a Biological Scientist II position in the laboratory of Dr. Shannon Boye. The research lab is developing viral vector-based gene therapies for the treatment of inherited ocular disease. The incumbent will perform laboratory experiments using gene therapy applications and procedures established in this lab. The individual in this position is expected to function under minimal supervision and troubleshoot simple to moderate complex scientific issues independently (minimally alter or modify procedures if necessary). The position requires work with rodent animal models, histological work, and molecular\/cellular work.<\/p>

Responsibilities to include:<\/p>

• Histological Work\/Analysis:<\/strong> Collecting organs and other tissues and embedding the specimens for sectioning either with a microtome or cryotome (for frozen sections). Following the preparation of slides with sections tissues, staining with standard histology stains such as H&E, as well as with immunohistochemistry will be done. Conduct image capture and analysis of tissue following intervention studies. <\/li>
• Molecular Techniques and Analysis:<\/strong> Molecular cloning, Plasmid purification, Primer design, RNA and DNA isolation, Isolating RNA and performing real-time PCR, and PCR genotyping of transgenic mouse lines; immunoassays to gene therapy vectors and transgenes, including ELISAs. This may include expansion\/maintenance of cell lines and analysis of transfection\/infection. Performing protein analysis by isolating protein of interest from cell or tissue lysates and performing\/analyzing western blots. Modifying protocols to improve existing techniques or to incorporate new diagnostic procedures.<\/li>
• Animal Studies:<\/strong> Handling mice for setting up timed matings, weaning animals and colony maintenance. Participating in treating animals with pharmacological agents or AAV vectors either through i.p., i.m., or i.v. injection. Other duties with the mice may include doing echocardiography, sensory and motor function, force mechanics, and metabolic studies. Plays a key role in regulatory management (IACUCs).<\/li>
• Regulatory Compliance:<\/strong> Ensures the laboratory is compliant with governing rules and regulations. Coordinate all IACUC and IRB (exempt) submissions\/renewals. Ensure all EH&S standards are met. Train new laboratory members to ensure compliance standards are met.<\/li>
• Training:<\/strong> Train and supervise graduate and undergraduate students performing diagnostic procedures related to gene therapy experiments.<\/li>
• Any other duties, as needed, to fulfill the mission and to abide by the values of the College of Medicine. This will include performing tasks and experiments as directed by the Principal Investigator.<\/li><\/ul><\/td><\/tr>

$34,000 - $44,000 annually; commensurate within this range based on qualifications and experience.<\/p><\/td><\/tr>

Bachelor\u2019s degree in biology or a closely related field and three years of relevant experience; or a Master\u2019s degree in biology or a closely related field and one year of relevant experience; Doctoral degree in biology or a closely related field.<\/p><\/td><\/tr>

• Experience in gene therapy research, specifically AAV<\/li>
• Animal colony management and handling of rodents experience.<\/li>
• Experience maintaining cell lines, performing plasmid transfections\/viral infections, immunohistochemistry, FACS, rt-PCR, genotyping, immunohistochemistry, western blotting, microscopy<\/li>
• Experience characterizing retinal function and structure in rodent models (ERG, OCT, fundoscopy, behavior testing), gene therapy and\/or virology are preferred.<\/li>
• Advanced degree in related scientific field.<\/li>
• Working knowledge of basic laboratory techniques.<\/li>
• Must be extremely knowledgeable in laboratory procedures and must be meticulous in recording, tracking & reporting of data collected.<\/li>
• Strong interest in translational research<\/li><\/ul><\/td><\/tr>
